Topical 8-bromo-cyclic GMP lowers intraocular pressure in rabbits.

چکیده

The topical application of 8-bromo-3'-5'-cyclic guanosine monophosphate (8BrcGMP) produces significant decreases in intraocular pressure (IOP) in rabbit eyes. Maximum effects are obtained with a 4% concentration, and IOP is reduced significantly (P less than 0.001) between 30 and 240 min after administration of the agent. The decrease in IOP occurs without significant change in tonographic outflow facility. A significant further decrease in IOP can be induced by topical 8BrcGMP in rabbits whose IOP has been lowered by the systemic administration of the carbonic anhydrase inhibitor acetazolamide.

Topical application of a cyclic GMP analog lowers IOP in normal and ocular hypertensive rabbits.

PURPOSE To determine whether a new cyclic GMP analog, 2'-O-(4-benzoyl)benzoylguanosine 3',5' cyclic monophosphate (BB-cGMP), significantly reduces intraocular pressure in either normotensive or hypertensive eyes of rabbits. METHODS Intraocular pressure of normal adult rabbits and/or rabbits with alpha-chymotrypsin-induced ocular hypertension was measured after topical application of BB-cGMP f...

Effects of l- and d-timolol on cyclic AMP synthesis and intraocular pressure in water-loaded, albino and pigmented rabbits.

Topical 2% l- or d-timolol reduced the elevation of intraocular pressure induced by water-loading in conscious rabbits. This drug effect appeared on the peak elevation (in pigmented eyes) and on the down-phase (in albino and pigmented eyes) of elevated intraocular pressure. The contralateral eye and the treated eye responded similarly. In urethane anesthetized, water-loaded rabbits, a greater i...

Increased intraocular pressure following topical azide or nitroprusside.

In rabbits the topical administration of sodium azide (NaNs) or sodium nitroprusside (SNP) increased intraocular pressure in a dose-response manner. These agents, which activate guanylate cyclase, elevated cyclic GMP in the aqueous humor. Systemic blood pressure and pulse were not altered. Tonographic outflow facility was unchanged, suggesting an increase in aqueous humor flow as the mechanism ...

Adenosine 3',5'-monophosphate analogue increases the outflow facility of the primate eye.

Our previous studies showed that cyclic-AMP increases the outflow facility of the eye of the rabbit. In this investigation, an analogue of cyclic-AMP, the 8-methylthio derivative, perfused into the anterior chamber of the eye of the vervet monkey, increased outflow facility twofold. This observation supports the hypothesis that cyclic-AMP mediates the action of adrenergic agents to reduce intra...

Effect of Calcium Channel Blockers on Intraocular Pressure in Rabbits

The objective of the study was to evaluate the antiglaucoma effect of calcium channel blockers diltiazem and verapamil. Albino rabbits were used and chronic glaucoma was induced in them using freshly prepared 150 units of alphachymotrypsin. 0.1 mL of drug solution was administered topically into the left eye whereas the right served as control. The pressure recording was carried out at 15, 30, ...
